ІР(3)-чутливі Са(2+)-канали ендоплазматичного ретикулума секреторних клітин зовнішньоорбітальних сльозових залоз щура

Встановлено роль інозитол-1,4,5-трифосфат (ІР3 )-чутливих Са2+-каналів у підтриманні Са2+-гомеостазу в секреторних клітинах сльозових залоз в умовах активації М-холіно- і Р2Y-рецепторів. Дослідження проведено на інтактних та пермеабілізованих секреторних клітинах зовнішньоорбітальної сльозової залози щурів. Клітини ізолювали за допомогою модифікованого нами методу Herzog, Sies, Miller (1976) та пермеабілізували дигітоніном (50 мкг/мл на 0,5 млн. клітин). Функціонування Са2+-транспортувальних систем оцінювали за зміною вмісту Са2+ у досліджуваних клітинах, який визначили спектрофотометричним методом із використанням арсеназо ІІІ.
Показано, що ІР3-чутливі Са2+-канали секреторних клітин сльозових залоз інгібуються 2-АФБ (10 мкМ) та активуються ІР3, а також за дії холіноміметиків (карбахолін) і агоністів пуринових рецепторів (АТР). За активації як М-холінорецепторів, так і Р2Y-рецепторів Са2+ вивільнюється з одного і того самого ІР3-чутливого депо, оскільки ефекти АТР і карбахоліну у високих концентраціях (1 мМ і 10 мкМ відповідно) є неадитивними. Збільшення вмісту Са2+ у клітинах внаслідок відновлення його концентрації в позаклітинному середовищі після попередньої мобілізації з депо карбахоліном чи тапсигаргіном підтверджує наявність депокерованих Са2+-каналів у секреторних клітинах сльозових залоз щурів.
