Стрес ендоплазматичного ретикулума, його сенсорно-сигнальні системи та роль у регуляції експресії генів за злоякісного росту і гіпоксії

Гіпоксія є одним з індукторів експресії великої групи генів, що контролюють гліколіз та процеси проліферації в умовах зниженого рівня кисню або внаслідок зменшеного його споживання. Разом із тим, гіпоксія є одним із чинників, що індукують стрес ендоплазматичного ретикулума, який, як і гіпоксія, є обов’язковим компонентом злоякісного росту і пов’язаний з цитоплазмою та ядром трьома сенсорно-сигнальними системами: PERK, ATF6 та ERN1. Виключення функції ERN1 – основного сенсорно-сигнального ензиму стресу ендоплазматичного ретикулума – призводить до зниження інтенсивності росту пухлин і змінює характер гіпоксичної регуляції експресії багатьох генів, що контролюють процеси проліферації та гліколізу. Сигнальний шлях ERN1 контролює експресію великої групи генів, залежних від стресу ендоплазматичного ретикулума та гіпоксії, і є важливим фактором росту злоякісних пухлин.
